研究目的 新規免疫関連転写制御因子であるZFAT(zinc-finger gene in AITD susceptibility region)を中心とした免疫系転写ネットワークを解明すること、そして免疫関連疾患発症におけるZFATの分子機序を解明することを目的とする。
研究概要 研究課題代表者が自己免疫性甲状腺疾患(AITD)感受性遺伝子として同定した新規免疫関連遺伝子ZFATは転写制御因子をコードすると考えられる。ZFATを軸とした①発現解析(マイクロアレイ・定量RT-PCR)、②タンパク-タンパク相互作用解析(Y2H, M2Hなど)、③ZFAT-DNA相互作用解析(ChIP法など)、によりヒト・マウス免疫担当細胞におけるZFATネットワークの全体像を明らかにする。
研究成果 ZFAT遺伝子は魚類からヒトに至るまで高度に保存された遺伝子であり、発生初期に不可欠な遺伝子であることが判明した。ZFATは正常T,B細胞およびヒト白血病細胞株で発現しており、細胞の増殖・分化およびアポトーシス制御機構に重要な役割を果たしていることが明らかとなった。タンパク質相互作用解析およびZFAT-DNA相互作用解析では、網羅的な基礎データの創出を行い、ZFATの転写制御に関するネットワーク解明への基盤を構築し、ZFATが細胞機能維持に重要な分子の制御に関与することが示唆された。
