研究目的 哺乳類の性は遺伝的に決定されるが、体細胞からの誘導をうけて、生殖細胞は自ら特異的な遺伝子カスケードを確立する。研究課題代表者が単離したNanos2遺伝子は雄の生殖細胞に特異的に発現しており、雄の生殖細胞の分化維持に必須である。ここでは、Nanos2の遺伝子カスケードを標的として、生殖細胞の性分化に関わるゲノムネットワークの解明を目指す。
研究概要 唯一次世代に受け継がれる生殖細胞の独自性の維持や、減数分裂の制御機構解明、インプリンティング確立など生殖細胞の性分化過程の解明をめざす。主にNanos2遺伝子ネットワークの解明を通じて生殖細胞性分化の理解と体細胞からのシグナルを伝達する機構を含めたシステムの解析を行なう。
研究成果 生殖細胞は体細胞から由来するレチノイン酸によって雌化する。一方、Nanos2はレチノイン酸の下流遺伝子群を抑制することにより減数分裂を阻害し雄化を進行させることが明らかになった。Nanos2は生殖細胞のRNAメタボリズ>ム制御(主にRNAの分解促進)を介して、多くの遺伝子発現に関与することが明らかになった。

実験情報 胎仔期生殖巣から抽出した total RNA よりWhole Mouse Genome (4x44K) および Low RNA Input リニア増幅&ラベル化キット Plus, one-color (Agilent) を用いてアレイデータを取得した。
