研究目的 免疫細胞による自己-非自己の識別機構の解明は免疫学の根幹をなす課題であり、自己免疫病発症機構の理解や治療法開発に貢献する。多くの抗原に対する免疫応答はT細胞に依存するが、その自己-非自己識別能力獲得の場である胸腺の微小環境形成に関与する遺伝子制御ネットワークを明らかにする。
研究概要 研究代表者は胸腺微小環境形成に、転写因子NF-κBファミリーの活性化を誘導するclassical経路とalternative経路によるシグナル伝達とそのクロストークが必須であることを明らかにしてきた。本研究課題では、胸腺の器官形成における両経路が形成するネットワークを時空間的に解析する。
研究成果 2種類のTNF受容体ファミリーの受容体が協調的にかつ時期特異的に機能することによりTRAF6とNIKを介してNF-κBを活性化し、胸腺上皮細胞の分化を誘導することを明らかにした。また、NF-κB活性化シグナルの標的遺伝子を網羅的に同定した。
