研究目的 骨格系の形成や維持を支える破骨細胞、軟骨細胞、骨芽細胞など運動器系細胞における遺伝子の転写制御およびシグナル伝達ネットワークを解明することで、骨粗鬆症、関節リウマチなどの運動器疾患治療への分子基盤を確立する。
研究概要 トランスクリプトーム解析やプロテオーム解析による網羅的な発現情報、タンパク質間相互作用情報、プロモーター解析結果等を利用して、運動器系細胞の分化において鍵となる遺伝子を抽出する。その後ノックアウトマウスを作成、解析することで、運動器の形成・維持・老化のメカニズムを転写制御およびシグナル伝達ネットワークの観点から明らかにする。
研究成果 破骨細胞分化のマスター転写因子NFATc1を中心とした転写制御ネットワーク機構(J Biol Chem, 2005;J Exp Med,2005)やTecファミリーキナーゼ(Cell, 2008)、カルモジュリンキナーゼ(Nat Med, 2006)によるシグナル伝達ネットワーク機構を解明した。一方、骨芽細胞におけるNFAT転写因子群の重要性とその制御機構も明らかにし(Nat Med, 2005)、骨格系におけるNFAT転写制御ネットワークに関する知見を集積した。また、関節リウマチなどの炎症性骨破壊疾患において、Th17細胞が産生するIL17が骨芽細胞のRANKL発現を誘導し、破骨細胞による骨破壊を促進するメカニズム(J ExpMed, 2006)や破骨細胞が産生するプロテアーゼ、カテプシンKが樹状細胞によるTh17細胞活性化サイトカインの発現を制御するメカニズムを解明する(Science, 2008)など、骨破壊疾患の治療法確立への新たな道を切り開いた。
