研究目的 高等生物個体の発生を担う転写因子群は、類似の分子群からなるファミリーを形成している。また、これらの転写因子群が機能するためには、転写因子とその活性を調節するシグナル分子群が一体となって機能的ネットワークを形成している。本研究では、様々な遺伝子改変方法が利用可能なマウスをモデル生物として、時空間的に使い分けられる転写因子群とシグナル伝達分子群のネットワークの解析を行う。
研究概要 近年大きな社会問題となっている糖尿病やがん等の疾患発症に関与していると予想されるb-ZIP型転写因子とTGF-βシグナル分子群に注目し、遺伝子改変マウスを用いて機能的ネットワークを同定し、それらの疾患発症における関与を解明する。
研究成果 遺伝子改変マウスを用いたこれまでの研究で、b-ZIP型転写因子MafAおよMafBが膵臓β細胞の分化とインシュリンの産生に必須であること、TGF-βシグナルの下流で機能するSchnurri-2(shn-2)が脂肪細胞の分化に関与することを明らかにした。また、TGF-βによる血管新生に関与する新規転写因子ネットワークと血管内皮細胞におけるSmad2, Smad3の機能について明らかにした。
