研究目的 四肢形成をモデルとし、組織や器官発生で機能する「遺伝子ネットワーク」を解明することによって、新しい疾患治療法や再生医療法の開発に不可欠な遺伝子情報の基盤を構築する。
研究概要 マウス胚の遺伝子発現をホールマウントin situハイブリダイゼーション法(WISH)によって可視化し、発生段階に応じた遺伝子発現を3次元的実体顕微鏡により分析した。特に転写関連遺伝子についてデータベース(EMBRYS)を構築、遺伝子ネットワークの中核となる転写因子群を同定した。転写因 子に依存した遺伝子発現の量的変化を網羅できるハイスループットスクリーニング法や、転写因子の結合するゲノム領域を検知できるチップオンチップ法などを組み合わせ、各転写因子によって発現が制御される遺伝子、転写因子の発現を制御する遺伝子を各々同定した。
研究成果 1,600の転写関連遺伝子の時空間的な遺伝子発現を網羅したEMBRYSデータベースを>完成させ、各組織、臓器の発生にかかわる遺伝子クラスターを同定した。さらに、これを基にしたシステマティックな解析から、骨格や筋肉発生における新たな遺伝子ネットワークを見いだした。
