研究目的 ゲノム上の遺伝情報は、転写因子等のタンパク質複合体により、高度に制御を受けている。ヒトの場合、30億塩基対にも及ぶ巨大DNAから必要な部分だけを読み取るために、転写制御は高い階層性をもって機能しているが、その詳細は不明である。現在の転写研究は従来「転写因子」として分類されてきたタンパクにのみ解析の重点が置かれ、遺伝子が制御を受けた結果産物である転写産物の変動に焦点が当てられている。しかし、真に転写制御を理解するには既存のタンパク機能の枠組みにとらわれず、ゲノム上の様々なタンパクーDNA相互作用を網羅的に明らかにしていき、全遺伝子産物量とタンパク結合位置の相関を解析する、ゲノム学的視点からの転写研究が必要とされている。
研究概要 本研究ではゲノム全体を余すことなく解析可能なDNAタイリングチップを用い、研究課題代表者らが開発した高精度ChIP-chip法(DNAチップを用い転写因子の結合位置を網羅的に特定する方法)を構築し、ゲノムネットワーク研究参加各機関に提供しつつ、転写制御因子の結合位置と全転写産物の網羅的かつ定量的同定を行い、転写制御機構の階層性と連携(レギュローム)を明らかにすることを目指す。
研究成果 現在までに、全染色体あるいはプロモーターを中心として、ヒストンH3、H4のアセチル化修飾、活性型、非活性型RNAポリメラーゼ、p53、核内受容体、Sox9、Hes1等の局在の増殖、刺激、分化過程でのダイナミクスをとらえることに成功した。特に従来、染色体分配に於いて必須の機能を果たしていると考えられていたコヒーシンタンパクがCTCFと共に、インシュレーターとして機能し、インプリンティング遺伝子の転写制御に機能していることを示した業績は高く評価された。
