研究目的 新しいDNAの合成技術を駆使して高精度かつ迅速遺伝子解析法の開発をおこない、ゲノムネットワークプロジェクトにおけるゲノム解析の技術的な発展を図る。
研究概要 今日、遺伝子の発現解析や遺伝子多型解析などの方法はすべて天然型の核酸プローブを用いているため、避けがたいミスマッチ塩基対形成による本来検出されるべきでない間違った遺伝子を検出してしまう大きな問題がある。本研究では、この問題を根本的に解決する天然塩基を凌駕する人工塩基の開発を検討し、これをDNAやRNA分子に組込んだ遺伝子検出プローブを開発する。
研究成果 独自に開発した塩基部保護DNA法により高純度のDNAプローブを多孔質ガラス(CPG)上で合成し、ケミカルライゲーションの技術を取入れた新規SNP検出法を開発した。また、天然塩基の塩基対形成能と塩基識別能を超えた人工塩基を組み込んだ保護DNAプローブ法を開発した。
